Works just fine, if you don't mind a tiny bit of slowness in opening.
Works better with USB 2.0. Make sure you have plenty of extra empty
space on the drive, don't try it with the flash drive almost filled
with files. FM needs at least as much space as the file size for temp
files, and could use more.
Just plug the drive in, and open the files as usual. Of course the
machine must have FM installed on it. If they don't have a licenced
copy, have them download a 30-day demo from filemaker.com.

I presume you meant run the application from a flash drive (Win or Mac)...
if win, it won't work since the registry entries don't match the app
location. It is possible to create a registry overlay that you can "install"
in a second on any win machine.
