we have a E:\ drive
if so how?

I think what your actually asking is 'can the default website have its home
directory on a different drive'?

The answer is yes. In IIS manager open the properties of the default
website and on the home directory tab change the local path. If you are
using anonymous access ensure the IUSR_ account has read access
to the folder.
